用计算机拟合法制作ALT标准曲线

摘要(Abstract):

用赖氏法重复测丙氨酸氨基转移酶(ALT)标准曲线10次,计算机作线性回归和曲线拟合。直线回归结果:酶活性在不高于97U的范围内,相关系数(R)=0999,斜率(b)=255×10-3,截距(a)=00286;而达到200U时,R=0986,b=190×10-3,a=0066,显著性检验两者之间差别为极显著。表明:97U以下线性较好但不过原点,大于97U之后开始明显偏离该线性。拟合后得一非线性曲线方程:吸光度=0959U/(24752+U)(R=0995)。转换为实用标准曲线方程为:U=24752吸光度/(0959-吸光度)。高值病理标本测定用该标准曲线可不必稀释而得到准确的结果。
The ALT standard curves were made ten times repeatly. The linear regression shown that correlation coefficient (R) is 0 999, slope (b) is 2 55 × 10 -3 ,interpret(a) is 0 0286 under 97 U of activity; and that R is 0 986, b is 1 90 × 10 -3 , a is 0 066 while up to 200 U of activity.Both difference is very singnificant (P<0 01).This suggests the linearity exists only under 97 U.A non-linear equation is obtained by computer imitation. A (absorbance) = 0 959 U / (247 57+U) or U = 247 52 A/(0 959-A) (R=0 995). On the formula, a accurate value in pathological serum specimen of higher ALT activity without dilution may be gotten by calculation.
