ICP-MS法检测贵州10种地道药材中重金属元素含量Determination of Heavy Metals in 10 Kinds of Traditional Chinese Medicine from Guizhou Province by ICP-MS

摘要(Abstract):
目的:检测10种贵州本地药材重金属的含量。方法:微波消解法处理10种药材样品,硝酸和过氧化氢作为消解体系,以锗(Ge)、铟(In)、铋(Bi)作为内标元素,通过ICP-MS仪同时定量10种药材中的铅(Pb)、镉(Cd)、汞(Hg)、砷(As)、铜(Cu)的含量。结果:各元素线性关系均良好(r>0.998),方法检出限为0.009~0.082μg/L,平均回收率为90.09%~99.16%,精密度和重复性的RSD值均小于5.0%;Cu含量为1.502~6.803 mg/kg,As为0.172~1.816 mg/kg,Cd为0.000~0.230 mg/kg,Pb为0.163~3.909 mg/kg,Hg为0.000~0.087 mg/kg,5种中药材明确需要控制的有害重金属元素的残留量符合现行《药用植物及制剂进出口绿色行业标准》、《中国药典》(2015版)、GB-14882-94《食品中放射性物质限制浓度标准》限量指标。结论:ICP-MS具有简便、快速、准确、灵敏度高的优点,可用于中药材中重金属的定量测定。
Objective: To detect the contents of heavy metals in ten kinds of traditional Chinese medicines from Guizhou Province. Methods: Samples were conducted by microwave digestion instrument,with HNO_3 and H_2O_2being the digestion acid system. Ge,In and Bi were used as internal standard elements,5 kinds of heavy metals( Pb,Cd,Hg,As,Cu) were determined by ICP-MS in ten kinds of traditional Chinese medicines. Results: The linear relation of each element was good( r > 0. 998).The detection limit was 0. 009 ~ 0. 082 μg / L,the average recovery was 90. 09% ~ 99. 16%,and the precision and repeatability of RSD values were less than 5%. Content of Cu was in the range of 1. 502 to 6. 803 mg / kg,content of As was in the range of 0. 172 to 1. 816 mg / kg,content of Cd was in the range of 0. 000 to 0. 230 mg / kg,content of Pb was in the range of 0. 163 to 3. 909 mg / kg and content of Hg was in the range of 0. 000 to 0. 087 mg / kg. Residual quantity of five kinds of harmful heavy metals( Pb,Cd,Hg,As and Cu) needed to control met the limit standards of the《Current green trade standards of importing medicinal plants》,《Chinese pharmacopoeia》( 2015) and GB- 14882- 94 of the《Concentration of radioactive material limit standard in food》. Conclusion: This method was simple,rapid,accurate,sensitive,and suitable for the determination of heavy metals in traditional Chinese medicines and providedbasis for its quality control.
