PNU通过PI3K/AKT信号通路上调星形胶质细胞HSP70的表达

摘要(Abstract):

目的:研究PNU对星形胶质细胞热休克蛋白70(HSF70)表达的影响,并探讨其机制。方法:分离新出生24 h内乳鼠大脑星形胶质细胞原代培养并鉴定,传1~3代待细胞成熟后分为正常对照组、PNU处理组,PI3K/AKT信号通路阻断剂(LY294002)处理组和PNU+LY294002处理组,正常对照组不做任何处理,PNU处理组用5μmol/L PNU分别处理6、12、18、24 h,LY294002处理组只加10μmol/L LY294002处理,PNU+LY294002处理组是先加10μmol/L LY294002处理2 h后、再加入5μmol/L PNU共同处理18 h,免疫印迹法观察各组星形胶质细胞内HSF70蛋白的表达。结果:与正常对照组相比,PNU处理组星形胶质细胞内HSP70蛋白的表达上调(P <0. 05);与PNU处理组比较,LY294002+PNU处理组星形胶质细胞内HSP70表达明显受到抑制(P <0. 01)。结论:PNU通过PI3K/AKT信号通路调控星形胶质细胞HSP70蛋白表达。
Objective: To study the effect of PNU on the expression of heat shock transcription factor1( HSF1) in astrocytes and its action mode. Methods: Primary rat astrocytes were isolated from newborn rats within 24 h,cultured and verified as astrocytes. After astrocytes became mature,they were divided into four groups: control group,PNU group,LY294002( PI3 K/AKT inhibitor) group and the PNU + LY294002 group. The control group was untreated; the PNU group was given 5 μmol/L PNU for 6 h,12 h,18 h and 24 h,respectively; LY294002 group was treated with 10 μmol/L LY294002;PNU + LY294002 group was pretreated with 10 μmol/L LY294002 for 2 h and then given 5 μmol/L PNU for 18 h. HSP70 expression levels were detected by Western Blot. Results: HSP70 level in PNU group was upregulated( P < 0. 05) when compared with the control group. LY294002 significantly inhibited PNU-upregulated HSP70 expression( P < 0. 01). Conclusion: PI3 K/AKT pathway mediates PNU-upregulates HSP70 expression in astrocytes.
