HPA1-5.15基因多态性与深静脉血栓形成的相关性

摘要(Abstract):

目的:探讨人类血小板抗原(HPA)1-5.15基因多态性与深静脉血栓(DVT)形成的相关性。方法:在贵州地区汉族人选择40例初发深静脉血栓形成患者为DVT组,另40例健康体检者40例为对照组,分别提取外周血基因组DNA,应用序列特异性引物-多聚酶链反应技术扩增目的基因,对HPA基因进行分型,采用Har-dy-Weinberg平衡检验HPA基因频率群体分布,χ2检验比较DVT组和对照组频数差异。结果:DVT组与对照组实际基因型分布与理论基因型分布差异无统计学意义(P>0.05),符合Hardy-Weinberg平衡定律,具有人群代表性;比较对照组和DVT组6个基因多态性差异无统计学意义(P>0.05)。结论:贵州地区汉族人HPA1-5.15基因多态性不是深静脉血栓形成的危险因素,与深静脉血栓形成无明显关系。
Objective: To explore the relationship of human platelet alloantigen(HPA)1-5.15 gene polymorphism with deep vein thrombosis.Methods: Forty Han nationality cases with incipient deep vein thrombosis(DVT)in Guizhou region served as DVT group and 40 healthy subjects served as control group.Genomic DNA was extracted from peripheral blood.Sequence specific primer-polymerase chain reaction was employed to amplify target genes.HPA gene was genotyped.Hardy-Weinberg equilibrium test was used to detect the population distribution of HPA gene frequency,and frequency differences between experimental group and control group were compared by using χ2 test.Results: No statistical difference was found in the practical and theoretical genotype distribution between DVT group and control group(P>0.05),which accorded with Hardy-Weinberg equilibrium law and showed a population representativeness.There was no statistical difference in polymorphism of 6 genes between control group and DVT group(P>0.05).Conclusions: HPA1-15 gene polymorphism is not a risk factor of DVT in Guizhou Han population.No significant relationship between HPA1-5.15 gene polymorphism and DVT is found.
