用抗弓形虫单克隆抗体检测孕妇血清中弓形虫循环抗原
Use of McAb Dot-ELISA to Detect Circulating Antigen of Toxoplasma Gondii in Serum of Pregnant Woman

摘要(Abstract):

用抗弓形虫单克隆抗体Dot-ELISA检测350例孕妇和18例娩出畸形儿的产妇血清中的弓形虫循环抗原(CAg),同时用Dot-ELISA检测这批血清中的弓形虫抗体IgG。350例孕妇血清中弓形虫CAg阳性3例,阳性率为086%。弓形虫IgG阳性18例,阳性率为514%。332份弓形虫IgG阴性血清CAg均为阴性。18例IgG阳性血清中3例CAg阳性,抗体滴度分别为1∶40,1∶40,1∶160,提示孕妇有早期活动性感染;还提示检测弓形虫IgG阳性抗体滴度不高时应作CAg检测,不可轻易作为既往感染以免漏诊了孕妇早期感染致使胎儿感染弓形虫。18例产妇血清中有2例血清CAg和IgG均为阳性,并分别从畸胎组织和脐血和羊水中分离出弓形虫。5例疟疾病人血清均无交叉反应。实验结果显示抗弓形虫单克隆抗体Dot-ELISA检测弓形虫CAg敏感性高、特异性强、方法简易、快速,是弓形虫病早期临床诊断的较理想方法。
The circulting antigen(CAg) of T. gondii(T.g) was detected in sera of 350 pregnant women and 18 lying in women who delevering deformed fetus. T. gondii monoclonal antibody (T.gMcAb) Dot ELISA was used to detect the CAg. Serum IgG was detected by Dot ELISA simultaneously. 3 of 350 pregnant sera were T.gGAg positive (0.86%),while 18 of 350 pregnant sera were T.gIgG positive (5.14%).In all of the 332 IgG negtive sera the CAg was negtive too. 3 of 18 IgG positive sera were CAg positive, the titer of IgG wese 1∶40,1∶40 and 1∶160 respectively. It indicated that the pregnant woman was in early active infection. It also showed that the CAg must be detected when the titer of IgG was low, in order to make a definite diagnosis of early infection. Because the early infection of pregnant woman may often cause fatal infection. The CAg and IgG were both positive of 2 in 18 lying in women's sera and the T.g were isolated from the deformed fetus. There was no oross reaction with 5 sera of malaria. The results showed that the T.gMcAb Dot ELISA is provided high sensitivity and sepcificity as well as the technic is simple to operate, it is an ideal method for early clinic diagnosis of toxoplasmosis.
