周期性压应力对成骨细胞增殖及活性的影响

摘要(Abstract):

目的:研究周期性压应力对成骨细胞增殖及活性的影响。方法:提取24 h新生SD大鼠成骨细胞并培养至第三代,鉴定后接种于六孔板,采用E1级饼形实验砝码加压(分别为0、1.42、2.83、4.25和5.66 g/cm2),压应力以加压30 min再停止加压30 min的方式循环进行,作用时间分别为3、6、9、12和24 h,设0 g/cm2压应力为对照组,1.42、2.83、4.25和5.66 g/cm2压应力为实验组;采用活细胞计数试剂盒(CCK-8试剂盒)及碱性磷酸酶(ALP)活性检测不同周期性压应力对成骨细胞生长增殖数目和细胞活性的影响。结果:经鉴定,原代培养细胞符合成骨细胞特性;周期性压应力作用6 h,1.42和2.83 g/cm2实验组成骨细胞OD值大于较对照组(P<0.05),但作用12和24 h时,前两者细胞OD值小于对照组(P<0.05);周期性压应力作用3 h,2.83 g/cm2实验组成骨细胞ALP活性较对照组及其他实验组高(P<0.05);作用9、12和24 h,各实验组成骨细胞的ALP活性均低于对照组(P<0.05);各时间点,4.25和5.66 g/cm2实验组的成骨细胞OD值及ALP活性均低于对照组及其他实验组(P<0.05)。结论:短时间内0~2.83 g/cm2的周期性压应力促进成骨细胞生长,但随着时间的延长,细胞增殖及活性受到抑制;2.83~4.25 g/cm2的周期性压应力对成骨细胞增殖有抑制作用;>4.25 g/cm2周期性压应力能抑制成骨细胞的增殖及活性。
Objective: To investigate the effect of periodic compression force on osteoblasts proliferation and Alkaline phosphatase activity. Methods: To extract primary osteoblasts from SD rats,cultivate them to the third generation and plant them into the 6-well plates,and then to conduct different periodic compression force with various experimental weight of level E1 for different periods of time,then preliminarily evaluate the effect of early osteoblasts proliferation on Alkaline phosphatase( ALP)activity by using cell counting kit-8( CCK-8) and ALP activity detection. Results: By conducting periodic compression force for 6 h,the osteoblasts proliferation of the 1. 42 g/cm2,2. 83 g/cm2 experimental groups was more than that of the control group( P < 0. 05). But when it was conducted for 12 h and 24 h,the osteoblasts proliferation in the two experimental groups was lower than that in the control group( P < 0. 05). By periodic compression force for 3 h,ALP activity of the 2. 83 g/cm2 experimental group was higher than the up-regulation as time went on,the osteoblasts proliferation rate and activity of the former were significantly higher than those of the control group and other experimental groups( P < 0. 05); The ALP activity of each experimental group with the compression force for 9 h,12 h and 24 h was lower than that of the control group( P < 0. 05). The osteoblasts proliferation and ALP activity of the 4. 25 g/cm2,5. 66 g/cm2 experimental groups were lower than those of the control group and other experimental groups( P < 0. 05). Conclusion: Present experimental conditions,the periodic compression force between 0 g/cm2 and 2. 83 g/cm2 promotes osteoblasts growth within a short time,but the osteoblasts proliferation and activity are inhibited as the time goes on. The periodic compression force between 2. 83 g/cm2 and 4. 25 g/cm2 may inhibite osteoblasts proliferation; The periodic compression force of over 4. 25 g/cm2 may severely inhibit cell proliferation and activity,and even lead to apoptosis.
