霉酚酸酯对大鼠肾间质纤维化的保护作用

摘要(Abstract):

目的:探讨霉酚酸酯(MMF)在肾间质纤维化模型大鼠中的肾脏保护作用及其可能的作用机制。方法:采用单侧输尿管结扎(UUO)致大鼠肾间质纤维化。将大鼠随机分为3组:假手术组、模型组、霉酚酸酯治疗组。手术后第9天处死各组大鼠,分别经免疫组织化学方法观察各组大鼠肾组织的血小板源性生长因子-B(PDGF-B)的表达部位、表达水平及α-平滑肌肌动蛋白(α-SMA)、增殖细胞核抗原(PCNA)的表达情况。M asson染色评定各组肾小管间质损害程度。结果:模型组PDGF-B,α-SMA,PCNA的表达及肾小管间质损伤指数明显高于假手术组(P<0.05),而霉酚酸酯组均明显低于模型组(P<0.05)。各项指标作相关分析,PDGF-B与肾小管间质损伤指数(r=0.870,P<0.01),α-SMA(r=0.956,P<0.01),PCNA(r=0.881,P<0.01)呈显著正相关。结论:PDGF-B在肾小管间质纤维化的病程进展中起重要促进作用,霉酚酸酯则可能通过下调PDGF-B的表达减轻肾小管间质纤维化。
Objective: To investigate the protective effects and its mechanism of Mycophenolate Mofetil(MMF) on rats with renal interstitial fibrosis.Methods: Rats were randomly divided into 3 groups: sham group(sham operated,SH),model group (disease models that did not receive MMF treatment,MD),and MMF treatment group(disease models that received MMF treatment,MT).Rat models of renal interstitial fibrosis were established by unilateral ureter obstruction(UUO).These animals were killed on the nineth day of the experiment,and the expression of platelet-derived growth factor-B(PDGF-B),α-smooth muscle actin(α-SMA) and proliferating cell nuclear antigen(PCNA) in the obstructed kidneys were measured by immunohistochemistry method.At the same time,the degree of tubulointerstitial damage was scored.Results: Compared with group SH,the levels of PDGF-B,α-SMA and PCNA in the group Md were significantly higher(P<0.05) and corresponded with the degree of tubulointerstitial damage.The levels of PDGF-B,α-SMA and PCNA in group MT were lower than those of group MD(P<0.05) due to the MMF treatment.The correlation coefficients of PDGF-B were r=0.870(P<0.01) with tubulointerstitial damage,r=0.956(P<0.01) with α-SMA,and r=0.881(P<0.01) with PCNA respectively.Conclusions: PDGF-B may play an important role in the development of renal interstitial fibrosis.MMF may prevent fibrosis development via down-regulating the expression of PDGF-B partly.
