摘要(Abstract):
目的:探讨过氧化物酶2(SOD-2)与沉默信息调节因子-3(SIRT3)在阿尔兹海默病(AD)患者不同脑区的表达,评估两者的相关性。方法:10例AD患者(AD组)和10例同龄正常人(对照组)尸体解剖后的脑组织,选择颞叶、额叶、海马和小脑脑区,应用免疫组织化学方法检测不同脑组织中SOD-2及SIRT3的分布和表达,采用Spearman分析在不同脑区SOD-2与SIRT3表达水平的相关性。结果:与对照组相比,AD患者颞叶、额叶、海马中SOD-2和SIRT3阳性细胞数及IOD值明显降低,差异有统计学意义(P<0.05);而在小脑中SOD-2及SIRT3表达较弱,且两组间比较,差异无统计学意义(P>0.05);AD患者的颞叶、额叶、海马中SOD-2与SIRT3的表达呈正相关(r~2=0.425,P=0.041;r~2=0.411,P=0.046;r~2=0.402,P=0.049),而在小脑中则无明显相关关系(r~2=0.031,P=0.624)。结论:AD患者大脑颞叶、额叶、海马中SOD-2和SIRT3表达水平降低,且两者呈现一定的相关性,可能与该病的神经病理学改变有关。
Objective: To investigate the expression of peroxidase 2(SOD2) and sirtuin-3(SIRT3) in different brain regions of patients with Alzheimer's disease(AD) and evaluate their correlation. Methods: Brains from ten patients with AD(AD group) and ten normal brain tissues with normal age(control group) were collected. The expression levels of SOD2 and SIRT3 were detected using immunohistochemistry in temporal lobe, frontal lobe, hippocampus and cerebellum. Spearman analysis was used to assess the association of the distribution and expression of SOD2 and SIRT3 in different brain regions.Results: Compared with the control group, the number of SOD-2 and SIRT3 positive cells and the IOD value were significantly lower in the temporal lobe, frontal lobe and hippocampus of patients with AD than those in control group(P<0.05). The expression of SOD-2 and SIRT3 were weak in the cerebellum, and there was no significant difference between AD group and control group(P>0.05). SOD2 expression levels were positively correlated with SIRT3 expression levels in temporal lobe, frontal lobe and hippocampus of AD patients(r~2=0.425, P= 0.041; r~2=0.411, P=0.046; r~2=0.402, P=0.049, respectively), while they were not significantly correlated in the cerebellum(r~2=0.031, P=0.624). Conclusion: The expression levels of SOD-2 and SIRT3 in the temporal lobe, frontal lobe and hippocampus of AD patients are decreased, and SOD2 was positively correlated with SIRT3. It may be related to AD neuropathological changes.
