生长抑素在海洛因依赖大鼠颌下腺的表达

摘要(Abstract):

目的:研究生长抑素(somatostatin,SS)在海洛因依赖大鼠颌下腺的表达。方法:正常SD大鼠,随机分为正常对照组、盐水对照组及海洛因依赖组,皮下注射海洛因建立海洛因依赖大鼠模型,分别于第10、17、24、31、38天取颌下腺,应用免疫组织化学SABC法及图像分析方法检测SS的表达。结果:SS阳性细胞数量及免疫反应强度在海洛因依赖第10天下降,之后随成瘾时间延长呈逐渐增强趋势;图像分析显示,SS阳性细胞数量在第31天时最高(P<0.05),平均灰度值在第31天时最低(P<0.05)。结论:海洛因依赖期间,生长抑素在大鼠颌下腺表达水平的变化可能是机体对海洛因依赖产生的一种适应性调节过程。
Objective:To observe the expression of somatostatin (SS) in submandibular gland of heroin dependent rats. Methods: Seventy SD rats were divided into normal control group (NCG,n=10),saline control group (SCG,n=30) and heroin dependence group (HDG,n=30). Heroin dependence model of rats were established by injecting with heroin subcutaneously. The immunohistochemical SABC method and image analysis were used to detect SS expression in submandibular gland on the 10th,17th,24th,31st and 38th days after the injection respectively. Results: The number of SS positive cells and staining intensity in submandibular gland decreased obviously in HDG,on the 10th day and then increased gradually along with the prolongation of heroin dependent time. The peak of SS positive cell number was on the 31st day showed by image analysis (P<0.05),while the lowest value (P<0.05) of mean grey degree was obtained on the same day. Conclusions: The changes of SS expression in rat submandibular gland during heroin dependence might be an adjusting process of the organism to injury caused by heroin.
