贵州安顺王锦蛇体内猬迭宫绦虫成虫及裂头蚴超微结构
Ultrastructure of Adult Spirometra erinacei and Pleroceroid within Elaphe carinata in Guizhou Anshun District

摘要(Abstract):

目的:观察贵州安顺王锦蛇体内猬迭宫绦虫成虫和裂头蚴的超微结构。方法:采集安顺地区王锦蛇体内的裂头蚴,进行形态学及分子鉴定;将裂头蚴喂饲感染犬(10条/犬)1月后剖杀,获得成虫,将成虫及裂头蚴标本用戊二醛-锇酸双重固定、乙醇梯度脱水、真空冷冻干燥及喷金后,用S-3400扫描电镜观察其超微结构。结果:(1)裂头蚴经cox1基因片段测序结果与Gen Bank中的猬迭宫绦虫(登录号GQ999947)的基因序列一致性为99.12%;(2)成虫体表微毛覆盖,背、腹面上各有一条纵行的吸槽,其内面的微毛呈丝状型,其余部位呈圆锥状棘状型;成虫头节呈指状、颈部短而粗,成节和孕节腹面有5条纵行的半球形颗粒状丘团,在成节腹面上部中央,从上而下,可见雄性生殖孔、雌性生殖孔及子宫孔,可见阴茎自雄性生殖孔伸出,阴茎呈短圆柱状,生殖孔周围由许多生殖乳突围绕;(3)裂头蚴呈长带状,前后两端略膨大,不分节,但表面有深的不规则的横向皱褶;虫体前端中央有一窄横向凹陷,周围体壁呈唇状突起,后端的结构类似前端,但中央的凹陷及唇状突起较前端扁平;虫体体表密布微毛,微毛的长短不一,前段及后段的微毛为圆锥状的棘状型,中段的微毛有圆锥状、棘状型和镶嵌在圆锥状微毛中矛尖状的棘状型,后者数量较少。结论:通过对贵州安顺王锦蛇体内猬迭宫绦虫成虫和裂头蚴的超微结构观察,有助于王锦蛇体内猬迭宫绦虫的后续研究。
Objective: To observe the ultrastructure of adult Spirometra erinacei and Pleroceroid within Elaphe carinata in Anshun District,Guizhou Province. Methods: Pleroceroids were collected from the wild Elaphe carinata in Anshun district,and were identified morphologically and PCR. The dogs were orally infected with Pleroceroids(10 Pleroceroids / each dog). After 4 weeks,infected dogs were sacrificed to observe and collect adult parasites. The Pleroceroids and adult parasites samples were fixed by glutaraldehyde osmic acid,dehydrated in graded series of ethanol,dried via vacuum freeze and coated with carbon gold. They were then observed by scanning electron microscopy( S-3400N).Results:(1) Results of gene sequencing of adult Pleroceroids had gene order consistency of 99. 12%with Spirometra erinacei in Gen Bank( Registration No. GQ999947);(2) adult parasite body was covered by microtriches,a longitudinal bothrium runs along the back and ventral side respectively; its interior microtriches were in silklike form of,the rest in cone shape spine-like form; scolex was in finger like shape,neck part was short and thick;five longitudinal papillaes run along ventral surface of mature proglottids and gravid proglottid venter. In the middle of mature proglottids had male gentital pore,female genital pore and uterus pore from top to bottom. Cylindroid cirrus could be seen protruding from the male genital pore in some mature proglottids,around which genital papilla were densely arranged.(3) The anterior and posterior ends of the Pleroceroids were slightly swell and without segmentation.Horizontal furrows ran irregularly along the body surface. The anterior end center was drawn inside which was a narrow dorsoventral hollow,around which,the plasma membrane displayed a lip bulge.Posterior structure was similar with the anterior one,just the drawn inside end center and lip bulge was flatter. The whole body surface of the Pleroceroids was densely covered by microtriches which differed in length. The microtriches of anterior and posterior segments were coniform spinithrix,while microtriches displayed two types( coniform spinithrix and lanceolate spinithrix) in the middle segment. Few lanceolate spinithrix were embedded in the coniform spinithrix. Conclusions: The studies of ultrastructure of adult Spirometra erinacei is benefitial for the future study of adult Spirometra erinacei and Pleroceroid within Elaphe carinata in Anshun District.
