中药头花蓼对幽门螺杆菌CagA及VacA表达的影响

摘要(Abstract):

目的:观察中药头花蓼对幽门螺杆菌(Helicobacter pylori,H.pylori)细胞毒素相关蛋白(cytotoxin-associated protein,Cag A)和空泡毒素(vacuolating cytotoxin,Vac A)表达的影响。方法:H.pylori ATCC700392接种于含头花蓼的哥伦比亚血平板上为实验组,同批未经药物作用的H.pylori ATCC700392为对照组,分别提取两组细菌的RNA和蛋白,用Real-time PCR定量检测头花蓼作用后cag A和vac A的mRNA表达,ELISA检测蛋白表达量。结果:经头花蓼作用的实验组与对照组相比,H.pylori的Cag A和Vac A蛋白表达水平分别降低了36.4%、55.1%(P<0.05),cag A和vac A的mRNA表达水平分别降低了69%、51%(P<0.05)。结论:头花蓼可能通过下调cag A和vac A的mRNA和蛋白的表达来发挥其对H.pylori的抑制作用。
Objective: To study the effect of traditional Chinese medicine Polygonum capitatum on expression of cag A and vac A of Helicobacter pylori( H. pylori). Methods: H. pylori ATCC700392 vaccinated in Colombia blood AGAR containing Polygonum captitatum was defined as experimental group,while the same batch H. pylori ATCC700392 without Polygonum captitatum as control group.After continuous and stable culture,bacterial RNA and protein were extracted respectively in two groups. Real- Time PCR was adopted to detect the mRNA expression of cag A and vac A,and ELISA was used to detect the protein expression of Cag A and Vac A. Results: Compared with control group,the protein expression of Cag A and Vac A in experimental group decreased 36. 4% and 55. 1%,respectively( P < 0. 05),while mRNA expression levels also decreased significantly,69% and 51%,respectively( P < 0. 05). Conclusion: Polygonum captitatum possibly exerts its inhibitory effect on H.pylori through inhibiting the expression of cag A and vac A.
