头花蓼对幽门螺杆菌尿素酶活性及基因表达的影响

摘要(Abstract):

目的:观察苗药头花蓼对幽门螺杆菌(H.pylori)尿素酶活性及尿素酶编码基因ure A、ure B表达水平的影响,探讨头花蓼抑制H.pylori的可能机制。方法:H.pylori分为对照组、1/2最低抑菌浓度(MIC)、1/4MIC、1/8MIC头花蓼处理组,分别于不含药的10%脱纤维羊血哥伦比亚血琼脂平板及含2、1、0.5 g/L头花蓼的10%脱纤维羊血哥伦比亚血琼脂平板培养3代,尿素酶活性比色法定量检测各组H.pylori尿素酶活性,Real-time PCR检测对照组、1/2MIC头花蓼处理组尿素酶基因ure A、ure B的mRNA水平。结果:头花蓼作用后,处理组H.pylori尿素酶活性均显著低于对照组,差异具有统计学意义(P<0.05),且头花蓼的浓度越大,H.pylori尿素酶活性受抑制的程度也越大;1/2MIC头花蓼处理后,H.pylori尿素酶编码基因ure A、ure B mRNA水平与对照组比较均显著下调,差异具有统计学意义(P<0.05)。结论:苗药头花蓼可通过抑制H.Pylori尿素酶活性及下调尿素酶基因ure A、ure B的表达水平抑制H.Pylori尿素酶的合成,从而降低H.Pylori在胃酸环境中的生存能力。
Objective: To observe the effect of Polygonum capitatum on Helicoba-cter pylori( H. pylori) urease activity and expression levels of urease gene ure A,ure B,and to discuss the possible mechanisms. Methods: H. pylori was divided into control group,treatment group by Polygonum capitatum with 1 /2 MIC,1 /4 MIC,1 /8 MIC,cultivated after three generations with defibrinated sheep blood Columbia blood agar base and same blood agar base contained 2,1,0. 5 g / L Polygonum capitatum concentration. Adopting Real-time PCR to detect the mRNA level of ure A,ure B of 1 /2 MIC and control group. Results: After processed by Polygonum capitatum,the H. Pylori urease activity of each treatment group were significantly lower than that in control group,the difference was statistically significant( P < 0. 05). The more concentrated the Polygonum capitatum was,inhibition level of such activity was greater; the mRNA level of urease gene ure A,ure B of 1 /2 MIC Polygonum capitatum was significantly down regulated compared with control group,the difference was statistically significant( P< 0. 05). Conclusion: The Polygonum capitatum can decrease the H. pylori viability in acid environment by inhibiting the urease activity of H. pylori and downregulating the urease gene encoding ure A and ure B to inhibit urease synthesizing.
