异汉防已甲素诱变性、致癌性筛检实验
A Screening Assay on the Mutagenicity and Carcinogenicity of Isotetrandrine

摘要(Abstract):

对贵州省抗矽肺新药——异汉防己甲素进行了诱变性、致癌性系列筛检实验。包括Ames试验、微核试验、姐妹染色单体互换(SCE)试验、精于畸形试验和显性致死试验,结果均为阴性。说明本品无致突变作用,致癌的可能性亦较小。
A series of sereening assays on the mutagenicity and carcinogenicity were performed. In Ames plate-incorporation assay, no mutagenic effects were found at the dose up to 500 μg/plate, in which the strains TA_(97) TA_(98) TA_(100) and TA_(102) were used. Isotetrandrine 225, 450, 900mg/kg per os did not induce any increment of micronucleus rate in mouse bone mallow cells. The frequencies of sister chromatid exchanges in mouse bone mallow cells in vivo were weakly positive at 120, 150mg/kg, and in a dosage of 18 or 54 μg/ml in the human peripheral blood lymphocytes cultured in vitro, it was found that the frequncies of SCE was increase significantly. For sperm malformation assay, administered 112.5, 225, 450 mg/kg for 5 days in male mice, the increased rates of abnormal sperm formation (5.6, 6.4, 5.2‰) were insignificant as compared to controls (3.0‰). The result of dominant lethal study showed that no increases of early fetal deaths and mutation index were noticed in female mice, which had matched with males, when adminis- tered 18, 60, 180mg/kg for 5 days.
