穴位埋线与糖通饮对早期糖尿病肾病大鼠血清TGF-β_1和IGF-1的影响
Effect of Catgut Embedding Therapy and Tangtong Drink on TGF-β_1 and IGF-1 Level of Early Diabetic Nephropathy Rats

摘要(Abstract):

目的:探讨穴位埋线、糖通饮防治早期糖尿病肾病(DN)的作用及机理。方法:60只SD大鼠随机分为正常对照组(12只)和造模组(48只),造模组大鼠采用腹腔注射STZ方法制作早期ND模型;造模成功后,随机分为模型组12只和治疗组(中药组、穴埋组、中药+穴埋组即药埋组,每组12只),根据分组进行相应的治疗4周,对照组和模型组灌胃生理盐水4周;疗程结束后72 h,剪尾取血测空腹血糖(FBG),检测血清和转化生长因子-β1(TGF-β1)及胰岛素样生长因子-1(IGF-1)水平,同时取大鼠肾组织计算肾脏肥大指数(KHI)。结果:治疗组(中药组、穴埋组、药埋组)大鼠较模型组大鼠FBG、TGF-β1、IGF-1血清水平含量显著降低(P<0.01),药埋组尤为明显;治疗组大鼠较模型组大鼠KHI明显降低(P<0.01)。结论:穴位埋线及糖通饮能有效降低早期DN大鼠TGF-β1、IGF-1血清水平含量,是防治早期DN的有效方法。
Objective: To preliminarily study effects and the possible mechanism of catgut embedding therapy and Tangtong drink on early DN rats. Methods: Sixty SD rats were randomly divided into the normal group( 12 rats) and the model group. The model group received intraperitoneal injection of STZ combined with high energy diet to establish early DN rats model. After model building,these model rats were divided into non-treatment group( 12 rats),and treatment group including Chinese medicine group,embedding group and the embedding plus Chinese medicine group,12 rats in each group. The treatment lasted 4 weeks and normal group and model group received intragastric administration of normal saline for 4 weeks. 72 hours after treatment,all rats were killed and the tail blood was collected to detect FBG level and serum TGF-β1,IGF-1level. Meanwhile,the rats' kidney tissue was taken to calculate the KHI. Results: Compared with non-treatment group,the FBG level and serum TGF-β1,IGF-1 level of the treatment group( Chinese medicine group,embedding group and the embedding plus Chinese medicine group) decreased significantly( P < 0. 01),of which,the embedding plus Chinese medicine group showed the most significant decrease. The kidney hypertrophy index( KHI) in rats of treatment group were significantly reduced compared with the non-treatment group( P < 0. 01). Conclusions: Catgut embedding therapy and Tangtong drink can reduce early DN rats'FBG level,TGF-β1level,IGF-1 level,and KHI. It is an effective method for the prevention and treatment of early DN.
