慢性氟中毒大鼠肝脏中P-糖蛋白和细胞核因子-κB的表达水平

摘要(Abstract):

目的:研究慢性氟中毒大鼠肝脏中P-糖蛋白(P-glycoprotein,P-gp)和细胞核因子κB(Nuclear factorκB,NF-κB)的表达变化,探讨氟中毒肝脏损害的发病机理。方法:将60只SD大鼠随机分为3组,即对照组、小剂量染氟组和大剂量染氟组,分别给予自来水、加10 mg/L和50 mg/L氟(Na F)的的自来水喂养6个月观察3组大鼠氟斑牙程度及尿氟,同时取大鼠肝脏;采用蛋白印迹方法检测P-gp和NF-κB的蛋白表达水平,实时荧光定量PCR方法检测NF-κB mRNA表达水平。结果:6个月后,小剂量染氟组、大剂量染氟组大鼠氟斑牙程度及尿氟含量明显高于对照组,差异有统计学意义(P<0.05);小剂量染氟组、大剂量染氟组大鼠肝脏中P-gp和NF-κB蛋白和mRNA表达高于对照组,差异有统计学意义(P<0.05);小剂量染氟组、大剂量染氟组NF-κB mRNA表达高于对照组,差异有统计学意义(P<0.05)。结论:慢性氟中毒引起大鼠肝脏中P-gp和NF-κB表达明显升高,这可能与氟中毒性肝脏损伤的发生机制有关。
Objective: To investigate the expressions of P-glycoprotein( G-pg) and nuclear factor κB( NF-κB) in the liver of chronic fluorosis rats,and to reveal the mechanisms of damaged liver resulted from the toxicity of the fluoride. Methods: The SD rats were randomly divided into three groups: control group,small dose fluoride exposure group and large dose fluoride exposure group. Three groups were fed with tap water,10 mg / L fluoride and 50 mg / L fluoride respectively for six months. Dental fluorosis degrees and urinary fluorine contents,and conditions of rats' livers were observed. The protein and mRNA expression levels of P-gp and NF-κB in livers were detected by Western blotting and quantitative real time PCR method. Results: Six months later,dental fluorosis degrees and urinary fluorine contents of small dose group and large dose group were remarkably higher than control group,differences were statistically significant( P < 0. 05); protein expression levels of P-gp and NF-κB in both fluoride exposure groups were significantly higher than those of control group,differences were statistically significant( P < 0. 05); NF-κB mRNA expression levels of both fluoride exposure groups were higher than those of control group,differences were statistically significant( P < 0. 05). Conclusions: The increased expressions of P-gp and NF-κB in the liver of rat are induced by chronic fluorosis,which is possible to correlate with the mechanism of liver damages triggered by fluoride poisoning.
