云南汉族人群HLA-DM基因多态性与HCV慢性感染的相关性分析

摘要(Abstract):

目的:研究云南汉族人群中HLA-DM基因多态性与丙型肝炎病毒(HCV)慢性感染的关系。方法:选取云南汉族人群HCV慢性感染者185人(病例组),健康对照180人(对照组),采用Sanger测序法和TA克隆测序法对HLA-DMA,HLA-DMB基因进行基因分型,计算这两个基因在病例组和对照组中的等位基因频率和基因型频率;构建单倍型,比较两组间的分布差异。结果:在病例组和对照组中,共检出4种HLA-DMA等位基因和5种HLA-DMB等位基因,DMA*01∶01-DMB*01∶03单倍型在病例组中的频率显著高于对照组(0.287 vs 0.197,OR=0.134,95%CI为1.529~2.305,P=0.005),DMA*01∶02-DMB*01∶03单倍型在病例组中的频率显著低于对照组(0.024 vs 0.056,OR=0.424,95%CI为0.194~0.944,P=0.031)。结论:单倍型DMA*01∶01-DMB*01∶03和DMA*01∶02-DMB*01∶03可能与云南汉族人群HCV慢性感染相关。
Objective: To investigate the association between HLA-DM gene polymorphism and chronic hepatitis C virus(HCV) infection in Yunnan Han population. Methods: We recruited 185 patients with chronic HCV infection(case group) and 180 healthy controls(control group) in Yunnan Han population. HLA-DMA and HLA-DMB genes were genotyped by Sanger sequencing and TA clone sequencing. Allele frequencies and genotype frequencies were calculated. In addition, haplotypes were constructed to compare the distribution frequency. Results: Four HLA-DMA alleles and five HLA-DMB alleles were detected in the both groups. The DMA*01∶01-DMB*01∶03 haplotype was significantly higher in the case group than that in the control group(0.287 vs 0.197,OR=0.134,95%CI was 1.529~2.305,P=0.005), while the DMA*01∶02-DMB*01∶03 haplotype was significantly lower in the case group than that in control group(0.024 vs 0.056, OR=0.424, 95% CI was 0.194~0.944,P=0.031). Conclusion: DMA*01∶01-DMB*01∶03 and DMA*01∶02-DMB*01∶03 haplotypes may be associated with chronic HCV infection in Yunnan Han population.
