HMGB1及MMP-9在非小细胞肺癌及癌旁组织中的表达水平

摘要(Abstract):

目的:分析高迁移率族蛋白B1(HMGB1)及基质金属蛋白酶-9(MMP-9)在非小细胞肺癌及癌旁组织的表达。方法:选取70例术后经病理组织切片确诊的肺癌患者的组织标本,根据病理学检查结果,将标本分为非小细胞肺癌组和癌旁组织组,鳞癌组和腺癌组,各病理分期组,采用组织化学方法观察各分组中HMGBI与MMP-9表达。结果:免疫组化结果显示,HMGB1及MMP-9在肺癌组织中具有较高的阳性表达率,HMGB1在非小细胞肺癌组织中的阳性表达率高于癌旁对照组,两组比较差异有统计学意义(χ2=11.15,P<0.05);从病理分型看,HMGB1在肺鳞癌组中阳性表达率明显高于腺癌组,差异有统计学意义(χ2=7.232,P<0.05);非小细胞肺癌组织中MMP-9的阳性表达率高于癌旁组织组,两组比较差异有统计学意义(χ2=21.39,P<0.05);从肺癌分期来看,Ⅰ~Ⅱ期中HMGB1阳性表达有27例,阳性率为64.2%;在Ⅲ~Ⅳ期中表达阳性的有23例,阳性率为82.1%;其中在Ⅰ~Ⅱ期中MMP-9阳性表达有25例,阳性率为59.4%;在Ⅲ~Ⅳ期中表达阳性的有21例,阳性率为75%。结论:HMGB1与MMP-9在肺癌组织中具有较高的阳性表达率,考虑HMGB1与MMP-9可能共同参与了肺癌的进展过程。
Objective: To investigate the expression of HMGB1 and MMP-9 in non-small cell lung cancer tissues and Precancerous lesions. Methods: 70 stored samples of thoracic resection surgery and histopathological slide diagnosed patients with lung cancer. The specimens were divided into two groups after pathology test,non-small cell lung cancer group( NSCLC group),precancerous lesions group,squamous cell carcinoma group,adenocarcinoma group and various pathological staging groups;adopting immunohistochemical method to observe the expression of HMGB1 and MMP-9 in all groups.Results: Analysis of experimental data indicated,HMGB1 and MMP-9 in lung cancer tissue has a relatively high positive expression rate. The positive expression rate of HMGB1 in non-small cell lung cancer was higher than precancerous lesions group,difference was statistical significant( χ~2= 11. 15,P < 0. 05); pathological analysis showed that the positive expression rate of HMGB1 in squamous cell carcinoma was higher than adenocarcinoma group,which was statistically significant( χ~2= 7. 232,P <0. 05). The positive expression rate of MMP-9 in non-small cell lung cancer was higher than precancerous lesions group,and the difference was statistically significant( χ~2= 21. 39,P < 0. 05). From lung cancer staging,positive expression rate in stage I to II was found in 27 cases,positive rate was64. 2%; 23 positive cases were found in stage III to IV,positive rate was 82. 1%; MMP-9 positive expression found 25 cases in stage I to II,positive rate was 59. 4%; 21 positive cases were found in stage III to IV,positive rate was 75%. Conclusion: HMGBI and MMP-9 have a high positive expression rate in lung cancer tissues,which proves that HMGB1 and MMP-9 are closely related to the development and progression of lung cancer.
