摘要(Abstract):
目的:探讨Toll样受体4(Toll like receptor 4,TLR4)mRNA及人β防御素2(human beta-defensin-2,HBD-2)在鼻黏膜组织中的表达及意义。方法:采用核酸分子原位杂交技术和免疫组织化学技术分别检测慢性鼻、鼻窦炎患者鼻黏膜组织中TLR4 mRNA和HBD-2的表达。结果:TLR4 mRNA在病变组鼻黏膜组织中的表达明显强于对照组(P<0.05)。但在病变组中13例Ⅰ型和17例Ⅱ型慢性鼻、鼻窦炎患者的病变鼻黏膜组织中的表达差异无显著性;HBD-2在对照组未见表达,而在病变组均出现不同程度的表达,两组间的表达差异具有显著性(P<0.01),而且在Ⅱ型病变鼻黏膜组织中的表达明显强于Ⅰ型(P<0.05)。结论:TLR4可能参与了鼻黏膜天然免疫,HBD-2可能是炎症反应的重要产物之一。致炎因子通过TLR4对病原微生物进行识别,激活鼻黏膜上皮细胞,最终导致炎性细胞产生大量的HBD-2,这可能在鼻黏膜启动有效的炎性免疫反应过程中具有一定的作用。
Objective: To investgate the expression and significance of TLR4 mRNA and HBD-2 in human nasal mucosa.Methods: Specimens from 30 patients with chronic rhinosinusitis and 15 noraml adults were examined by in situ hybridization for TLR4 mRNA and by immunohistochemistry for HBD-2.Results: All of the 30 samples of chronic rhinosinusitis showed a stronger expression of TLR4 mRNA(P<0.05) and HBD-2(P<0.01) than those of noraml adults.Conclusion: Epithelia in human nasal mucosa could present a novel defense mechanism during bacterial infections with the help of TLR4,and HBD-2 might be the product of the process.
