脐带血基础免疫物质分析
A Study of Basic Immunological Substances in Foetus Cord Blood

摘要(Abstract):

目的:通过胎儿脐带血免疫状况的研究,为脐带血有形和无形成分在免疫方面的应用提供理论依据。方法:应用血液常规检验、试剂盒以及液体闪烁计数仪和流式细胞仪对白细胞分类、免疫球蛋白、细胞因子、脐血T细胞抗原表面标志和脐血有核细胞NK活性进行测定。结果:与成人外周血中免疫成分相比,胎儿脐带血中IgA和IgE含量明显减少,IgG无明显改变;脐血浆中G-CSF和GM-CSF分别为成人外周血的11.37倍和15.81倍,TNF略低于成人外周血;中性粒细胞接近成人外周血低值,中、晚幼粒细胞数明显低于成人外周血,淋巴细胞与成人外周血相当,NK细胞活性亦略低于成人外周血;脐带血T细胞亚群CD3+、CD4+和CD8+细胞数低于成人外周血,而CD34+和CD45RA细胞则高于成人外周血。结论:脐血中富含大量的G-CSF和GM-CSF等相关造血因子和原始的造血干细胞成分,虽然淋巴细胞的发育多处于幼稚期,但大量相关的免疫细胞和细胞因子均可作为免疫治疗的基础物质。
Objective: To study the immunity state of foetus cord blood,so as to provide theorty basis for the application of immunological substances from foetus cord blood in hematoplasma immunotherapy.Methods: Using blood routine test kits,liquid scintillation counter and flow cytometry to detect leukocyte classfication,immune globulin,T cell antigen surface marker and NK cell respectively.Results:IgA and IgE in cord blood were much less than in adult peripheral blood,but IgG was nearly the same as that in adult peripheral.G-CSF and GM-CSF in cord blood plasma were 11.37 and 15.81 times respectively of those in adult peripheral blood,while tumor necrosis factor(TNF) was a little bit lower,and heterophil granulocyte level was close to the lower limit in adult peripheral blood.Metagranulocyte and myelocyte levels were obviously lower than those in adult peripheral blood.Leukomonocyte level was about the same as that of adult.The counts of subpopulations CD3+,CD4+,CD8+ of T cell in cord blood were significantly lower than those in adult peripheral blood while those of CD34+ and CD45RA were higher.NK cell killing activity was slightly lower than that of adult blood.Conclusion: Relative hemopoiesis factors and native hematopoietic stem cell components,G-CSF and GM-CSF,are abundant in cord blood and can be widely used in clinical immunotherapy,even though most T lymphocytes in cord blood are in naive stages of development.
