围绝经期综合征渐衰动物模型选择
Selection of Fading Animal Models with Perimenopausal Syndrome

摘要(Abstract):

目的:复制围绝经期大鼠动物模型,为围绝经期的实验研究奠定基础。方法:选取3月龄雌性未孕SD大鼠60只,分为假手术组和去势组;假手术组仅切除卵巢周围少量脂肪组织,去势组均切除右侧卵巢,去势1组、2组分别保留左侧卵巢体积约50%、20%,去势3组双侧卵巢全切,每组各15只;术后第7天行阴道脱落细胞涂片检查脱落细胞中角化细胞指数(CI),第21天取腹主动脉血检测血清雌二醇(E2)和卵泡刺激素(FFSH)水平,并取各组子宫组织观察病理变化。结果:假手术组、去势1、2、3组大鼠阴道涂片角化细胞CI分别为>50%、<50%、<30%和0,与假手术组相比,各去势组大鼠血清E2含量显著降低,血清FSH的含量显著升高(P<0.01),提示各去势组造模成功;去势1组E2水平最高,去势2组次之,去势3组最低(P<0.01);去势3组FSH水平最高,去势2组次之,去势3组最低(P<0.01);子宫组织形态学显示,与假手术组相比,各去势组大鼠子宫内膜上皮呈不同程度萎缩变薄,间质、腺体稀疏,血管少,其中以去势3组大鼠子宫萎缩最明显。结论:采取手术去势方法成功复制了围绝经期综合征渐衰大鼠动物模型,可大致模拟人类卵巢功能渐衰的过程。
Objective: To provide a theoretical basis for the experimental research and treatment of perimenopausal syndrome by copying perimenopausal animal models and discuss the feasibility for the treatment of perimenopausal syndrome. Methods: 60 female Sprague-Dawley( SD) rats of three months old were selected and divided into the control group and three castration groups( resected of the right ovarian),with 15 rats in each group. In Castration Group I,50% of the left of ovarian tissues of each rat was conserved; in Castration Group II,20% conserved and in Castration Group 3,all ovarian tissues of both sides were resected. 7 days after resection cornification index( CI) of vaginal exfoliated cells were examined. 21 days after resection,serum estradiol( E2) and follicle-stimulating hormone( FSH) were detected and the morphology change in uterine tissues was observed. Results: CI in the control group and Castration Group I,II and III were > 50% 、< 50% 、< 30% and 0 respectively Compared with the control group,the content of E2 of the castration groups was significantly lower the content of FSH in serum significantly higher,and the difference was statistically significant( P < 0. 01);Compared with the castration group,Castration Group I showed the highest estradiol and Castration Group II,the second,and Castration Group III the lowest; But Castration Group III showed the highest FSH and Castration Group II came to the second,Castration Group I the lowest,and the difference of FSH was statistically significant( P < 0. 01); According to the Uterine Morphology,compared with the control group,the thinner endometrial epithelial,interstitial glands and less blood vessels of all the 3 castration groups were observed. The rats' uterine contraction of Castration Group III was the most obvious. Conclusions: The surgical castration method successfully copied the menopausal animal models of rats perimenopausal syndrome,providing the similar evidence for the human ovarian aging process and for the early intervention of human perimenopause syndrome.
