灭蚊真菌卡地腐霉产生游动孢子所需条件的研究
A Study on Effects of Several Environmental Factors on Sporulation of Pythium carolinianum, a Fungal Pathogen of Mosquito Larvae

摘要(Abstract):

目的:观察卡地腐霉产生游动孢子所需的环境条件。方法:以卡地腐霉经诱导产生的游动孢子数量为指标,观察不同条件对其产生游动孢子的影响,确定该真菌产生游动孢子所需的最适条件。结果:卡地腐霉产生游动包子的适宜温度范围为10~30℃,最适温度为 24~26%;适宜产孢的蒸馏水pH值范围为5~12,最适pH值为9~11;8 d的培养物产孢量最多;菌块与蒸馏水的比例为1:40时产孢最多;三种光照周期对卡地腐霉的产孢影响不大,紫外线对产孢有抑制作用;NaCl、(NH_4)_2SO_4、蛋白胨和葡萄糖对游动孢子的产生有较强抑制作用,产孢能力对尿素的耐受性极大。结论:本观察所得资料对卡地腐霉的基础研究和应用都有重要价值。
Objective: To investigate the suitable environmental conditions needed by Pythium caro-linianum for sporulation. Methods: The amount of zoospores produced by P. carolinianum under different conditions was chosen as the index to reflect the suitability of the fungus to the conditions. Results: The suitable temperature range for sporulation of the fungus were 10 ℃-30℃, while the optimal temperature range was 24 ℃ - 26 ℃ ; The Suitable pH range was 5 - 12, with an optimum range of 9 - 11; Mycelia of 8 - day old had the highest zoospore yield; The optimal ratio of hyphal pieces to water volume(ml) was 1:40(10:400) for zoospore production; The light cycles did not show influence, but ultraviolet had obvious inhibitory effect on sporulation; Zoospore production was significantly reduced by NaCl, (NH4)2SO4, pepton and glucose at the concentrations of 0.2g/l, 50mg/l, 10mg/l, and 10mg/l respectively, but zoospore could be produced when high concentration of urea solution existed. Conclusions: This research has revealed the requirement of P. carolinianum to environmental conditions for sporulation. Data collected here are useful reference for further basic study and application of P. carolinianum.
