地黄饮子对急性脑缺血大鼠的学习记忆能力的影响

摘要(Abstract):

目的:探讨地黄饮子对急性脑缺血大鼠的学习记忆能力影响。方法:选取Wistar大鼠72只随机均分为实验1组、实验2组、模型组和正常对照组,前3组大鼠复制急性脑缺血模型,正常对照组大鼠手术但不给予脑缺血;实验1组造模后每日灌服地黄饮子汤36 g/kg,实验2组造模后每日灌服尼莫地平12.5 mg/kg,模型组和正常对照组造模后1次/d盐水灌胃,各组大鼠连续灌胃3周;所有大鼠进行Morris水迷宫定位航行实验记录逃避潜伏期测试学习能力,跳台实验记录逃避潜伏期评价记忆能力,并用电镜图像分析系统计算4组大鼠脑梗死面积。结果:与正常对照组比较,模型组大鼠定位航行实验潜伏期明显延长,差异有统计学意义(P<0.05);与模型组比较,实验1组和实验2组大鼠的定位航行实验潜伏期明显缩短,差异有统计学意义(P<0.05);与正常对照组比较,模型组大鼠跳台实验潜伏期明显延长,差异有统计学意义(P<0.05);与模型组比较,实验1组和实验2组大鼠的跳台实验潜伏期明显缩短,差异有统计学意义(P<0.05);实验1组脑梗死面积明显小于模型组和实验2组,差异有统计学意义(P<0.05)。结论:地黄饮子可以提高脑缺血再灌注大鼠的学习能力和记忆能力,减少缺血对脑组织神经元的损伤。
Objective: To explore the effect of Rehmannia Yin Zi on learning and memory ability of acute cerebral ischemia rats. Methods: 72 Wistar rats were randomly divided into experimental group1,experimental group 2,model group and normal control group. The acute cerebral ischemia model was established in the first 3 groups of rats while the normal control group did not received cerebral ischemia surgery. Experiment 1group received daily intragastric administration of Dihuangyinzi Decoction(36 g /kg) after modeling,experimental group 2 received daily intragastric administration of nimodipine(12. 5 mg / kg) after modeling,and model group and normal control group received 1 / d saline after modeling. All rats were administered orally for 3 consecutive weeks. All rats were subjected to Morris water maze navigation experiment to for memory capacity evaluation. Electron microscope image analysis system was adopted to calculate cerebral infarction area for the 4 groups of rats. Results: Compared with normal control group,the experimental latency of model group was significantly prolonged,and the difference was statistically significant(P < 0. 05). Compared with model group,the experimental latency in experimental group 1 and experimental 2 were significantly shortened,the difference was statistically significant(P < 0. 05). Compared with normal control group,the step- down test latency of rats in model group was significantly increased,and the difference was statistically significant(P <0. 05). Compared with model group,the step-down test latency of experimental group 1 and experimental group 2 rats were significantly shorter,and the difference was statistically significant( P <0. 05); The cerebral infarction area in experimental group1 was significantly smaller than the that of model group and experimental group 2,and the difference was statistically significant( P < 0. 05).Conclusion: Rehmannia Yin Zi can improve learning and memory ability of cerebral ischemia reperfusion rats,and reduce the damage of brain neurons resulted from ischemia.
