通络刮痧和中药对气滞血瘀证雌性大鼠肝组织Nrf2表达影响的对比研究Comparison of the Effects of Tongluo Scrapping and Traditional Chinese Medicine on Liver Nrf2 Expression of Female Rats with Qi-Stagnation and Blood-Stasis Syndrome

摘要(Abstract):
目的:探究通络刮痧和中药对气滞血瘀证雌性大鼠肝组织核因子E2相关因子2(Nrf2)表达的影响。方法:40只SD雌性大鼠随机均分为空白组、模型组、中药组及刮痧组,模型组、中药组及刮痧组大鼠连续14 d腹腔注射盐酸肾上腺素、4 h后接受1~2种不可预见性物理刺激制作气滞血瘀证动物模型,空白组大鼠不予注射、正常喂养;造模成功后次日,中药组大鼠用桃红四物汤灌胃每日1次、共治疗14 d,刮痧组大鼠给与通络刮痧法每周1次、共治疗2次;模型组大鼠每日抓捕1次、持续2周,空白组大鼠不予任何治疗、正常喂养2周;2周后,4组大鼠取血检测全血黏度(低切、中切及高切)、血浆黏度及血细胞比容,检测凝血酶时间(TT)、部分活化凝血活酶时间(APTT)、凝血酶原时间(PT)及纤维蛋白原(FIB),剖腹取出部分肝组织后,用玻璃匀浆器研磨均匀,检测肝组织Nrf2 mRNA及蛋白的表达。结果:与空白组相比,模型组大鼠血液黏度、血细胞比容都明显升高(P<0.05或P<0.01),TT和APTT明显缩短(P<0.05或P<0.01),肝组织Nrf2 mRNA及蛋白表达增强(P<0.05或P<0.01);与模型组相比,中药组和刮痧组大鼠血液黏度、血细胞比容都明显降低(P<0.05或P<0.01),TT及APTT延长(P<0.05或P<0.01),肝组织Nrf2 mRNA及蛋白表达显著增强(P<0.05或P<0.01);刮痧组和中药组大鼠各指标比较差异无统计学意义(P>0.05)。结论:通络刮痧和桃红四物汤可改善气滞血瘀证雌性大鼠部分血液流变学及凝血指标,其作用机制可能与上调肝组织Nrf2 mRNA及蛋白的表达有关。
Objective: o investigate the effects of Tongluo scraping and Chinese herbs on the expression of liver nuclear factor E2 related factor 2(Nrf2) in female rats with qi stagnation and blood stasis syndrome. Methods: Forty female SD rats were randomly divided into blank group, model group, traditional Chinese medicine(TCM) group and scraping group. Rats in the model group, Chinese medicine group and scraping group were intraperitoneally injected with adrenaline hydrochloride(Adr) for 14 days and given1~2 types of unforeseen physical stimulation to generate the animal model of qi stagnation and blood stasis syndrome. On the next day after successful modeling, TCM group was treated with Taohong Siwu Decoction once a day for 14 days, and the scraping group was given Tongluo scraping therapy once a week; the model group were given arrest once a day for 2 weeks. The blank group was given normal feeding. We examined blood viscosity(low cut, medium and high cut), plasma viscosity and hematocrit, four thrombin indicators including thrombin time(TT), partial activated thromboplastin time(APTT), prothrombin time(PT) and fibrinogen(FIB). Liver Nrf2 mRNA was assayed by qPCR and Nrf2 protein was detected by Western Blot. Results: Compared with the blank group, blood viscosity, hematocrit and the expression levels of liver Nrf2 mRNA and protein in the model group were significantly increased(P<0.05, P<0.01, P<0.05 and P<0.01, respectively), while TT and APTT were significantly shortened(P<0.05 and P<0.01, respectively). Compared with the model group, blood viscosity and hematocrit in TCM group and the scraping group were significantly decreased(P<0.05 and P<0.01), while TT and APTT were prolonged. There was no significant differences in the indexes between scraping group and TCM group. Conclusion: Tongluo scraping and Taohong Siwu Decoction can improve the blood rheology and coagulation parameters of female rats with qi stagnation and blood stasis syndrome. The mechanism may be related to the up-regulation of liver Nrf2 mRNA and protein expression.
